Ironically, being a member of DOMAIN\Domain Admins guarantees nothing.
In this case, verify that DOMAIN\Domain Admins is, in fact, a member of the
BUILTIN\Administrators group on the WSUS Administrator. I know that it's a
common practice to remove DOMAIN\Domain Admins from that group, and it's not
uncommon for this to be the cause of this particular behavior.

Of course, this additional information is useful, and could indicate a
number of other issues with accounts and/or permissions, including
unexpected changes to permissions or passwords on the IUSR_Machinename
account, the IWAM_Machinename account, the NTAUTHORITY\Network Service
account, or the NTAUTHORITY\Local System account - any of that sound
familair?

Could also be that the database service is stopped. Have you verified that
the database server is accessible, in general?
